II. Mechanism of Action
Targeting the interaction between FOXO4 and p53 proteins, it achieves selective serolysis of senescent cells. The core pathway is as follows:
Target Specificity: Senescent cells highly express FOXO4, which binds to and impounds p53 in the cell nucleus, preventing p53 from initiating apoptosis, maintaining the survival of senescent cells, and secreting SASP inflammatory factors (IL-6, TNF-α, etc.).
Competitive Blocking: After entering cells, FOXO4-DRI binds with high affinity to the TAD2 domain of p53, competitively disrupting the FOXO4-p53 complex and releasing p53.
Selective Apoptosis: Free p53 translocates to mitochondria, activating pro-apoptotic proteins such as BAX, inducing apoptosis only in senescent cells, with no significant effect on healthy proliferating cells.
Structural Advantages: D-type amino acid modification makes it resistant to in vivo protease degradation, resulting in a longer half-life and higher bioavailability.
